Abstract

This protocol describes methods used by the Editorial Office in the University of Edinburgh for in situ hybridisation and immunohistochemistry data entries for GUDMAP1, GUDMAP2 and nGUDMAP.

Procedure

The in situ hybridisation and immunohistochemistry data in the GUDMAP database complies with the specifications set out by MISFISHIE (Minimum Information Specification For an In Situ Hybridisation or Immunohistochemistry Experiment).

The following information is requested in a GUDMAP in situ hybridisation entry:

  • Gene symbol
  • Probe name (of the antisense riboprobe)
  • Clone name (used to generate the antisense riboprobe)
  • Sequence accession ID (describing the cDNA sequence of the antisense riboprobe)
  • Label (e.g. digoxigenin, FITC etc.)
  • Visualisation method (fluorescence, colorimetric)
  • Fixation method
  • Strain
  • Sex
  • Theiler stage
  • Genotype (if non-wildtype this must be specified)
  • Annotation (describing the pattern of gene expression)
  • Images (supporting the annotation)

The following information is requested in a GUDMAP immunohistochemistry entry:

  • Gene symbol (of the epitope recognised by the primary antibody)
  • Clone name (of the primary antibody)
  • Antibody type (monoclonal, polyclonal)
  • Supplier (the company that manufactured the antibody)
  • Catalogue number / Lot number (if a commercially available antibody)
  • Label (e.g. digoxigenin, FITC etc.)
  • Visualisation method (fluorescence, colorimetric)
  • Fixation method
  • Strain
  • Sex
  • Theiler stage
  • Genotype (if non-wildtype this must be specified)
  • Annotation (describing the pattern of gene expression)
  • Images (supporting the annotation)
